Descrição e referências: Alpha-GSU (Cga) knockout mice, with a neomycin resistance gene in exon 3, were generated by homologous recombination in embryonic stem cells and blastocyst injection at University of Michigan (Kendall et al., 1995). Chimeras were mated with C57BL/6 mice to produce a line on a mixed genetic background that are officially known as B6;129S2-Cgatm1Sac. Mice were maintained at University of Michigan and University of São Paulo, and they are deposited in The Jackson Laboratory (Stock number 002494). Mice were genotyped using the original protocol (Kendall et al., 1995).

Características (fenótipo): The CD40L- (or CD154-) knockout mutation is located on the X chromosome. Homozygous females and hemizygous males are viable and fertile. Percentages of B and T cell subpopulations are normal. Homozygous females and hemizygous males exhibit selective deficiencies in humoral immunity (low basal serum isotype levels and undetectable IgE) as well as abnormal secondary antigen-specific responses to immunization with a thymus-dependent antigen. The phenotype of the mice resembles human X-linked hyper IgM syndrome.

Características (fenótipo): Mice homozygous for the Ldlrtm1Her mutation have an elevated serum cholesterol level of 200-400 mg/dl and they have very high levels (>2,000 mg/dl) when fed a high fat diet. Normal serum cholesterol in the mouse is 80-100 mg/dl.
Descrição e referências: The Ldlrtm1Her mutant strain was developed in the laboratory of Dr. Robert Hammer and Dr. Joachim Herz at the Howard Hughes Medical Institute Research Laboratories, University of Texas Southwestern Medical Center at Dallas. The 129-derived AB1 ES cell line was used. The strain has been backcrossed to C57BL/6J mice for 10 generations (N10).

Características (fenótipo): Adenocarcinomas arise in virgin and breeder females and occasionally in males, which are well-differentiated, multifocal and eventually involve the entire mammary fat pad. Tumor-bearing females have reduced lung metastasis compared to the 80-90% incidence found in FVB/N-Tg(MMTV-PyVT)634Mul/J mice. Transgene expression is detected at high levels in male and female mammary glands. Lower levels are detected in salivary gland, seminal vesicles, ovaries, and lungs (believed to be the result of pulmonary metastases).

The resulting male mice heterozygous for the Prkdcscid allele and hemizygous for the Il2rgtm1Wjl allele were crossed to female NOD.CB17-Prkdcscid/J (Stock No. 001303) mice for eight generations. Heterozygotes were interbred to produce mice homozygous for the Prkdcscid allele and homozygous (females) or hemizygous (males) for the Il2rgtm1Wjl allele.

Mice were originally obtained from the Jackson Laboratory (DW/J Mlphln, Pou1f1dw/J, Stock number 000643) and maintained at University of Michigan and University of São Paulo. Mice were genotyped by Sanger sequencing using the primers 5’GCT GCT AAG GAT GCT CTG G 3’ and 5’ CCT TGG AAA TAG AGA ACA GGC 3’ and 5’ ACA CTT CGG GGA CCC CAG CA 3’ and 5’ CTC TGC CTT CGG TTG CAG GA 3’.

Descrição e referências: Ames dwarf mice, Prop1df/df are homozygous for the spontaneous mutation p.S83P, c.475C>T (ENSMUST00000051159). Mice were obtained from Dr. Andrez Bartke (DF/B) and maintained as a non-inbred, mixed stock at University of Michigan and University of São Paulo. Similar mice are available from the Jackson Laboratory (Oca2p, Prop1df/J; Stock number 001618). Mice were genotyped by PCR using the primers 5’ GAG CTG GGG AGA CCT AAG CTT TGC C 3’ and 5’ GCC CAG ATG TCA GGA TAC TG 3’, followed by PCR product digestion with Van91I.

Descrição e referências: The breeder pair of C57BL/10ScN mice, obtained from NIH, that were the progenitors of all mice of this strain at The Jackson Laboratory were tested by PCR analysis for the spontaneous Il12rb2 mutation described by Poltorak et al. (J. Immunol. 167:2106-2111, 2001) and found to carry only the wild type Il12rb2 allele. C57BL/10ScN mice have a deletion of the Tlr4 gene that results in absence of both mRNA and protein and thus in defective response to LPS stimulation. Tlr4lps-del differs from the Tlr4Lps-d mutation of C3H/HeJ mice, a point mutation that causes an amino acid substitution. The allele for normal LPS response, Tlr4lps-n, occurs in most other C3H and C57BL/10 substrains and most other mouse strains.
